5-Deazaflavin (TND-1128)

5-Deazaflavin, also known by research code TND-1128, is a synthetic analog of riboflavin (vitamin B₂) where the N-5 position of the isoalloxazine ring is replaced by carbon. This compound exhibits a unique auto-redox cycling ability – it can readily oscillate between oxidized and reduced states without enzymatic assistance. TND-1128 has been investigated as a mitochondrial energizer and neuroprotective agent. In recent pharmacological studies, low doses of TND-1128 activated mitochondrial function in brain neurons, improving their calcium handling and resistance to stress. Specifically, in a mouse model, TND-1128 pretreatment preserved neuronal mitochondrial membrane potential and prevented calcium overload during excitotoxic challenge, effects comparable or superior to those of β-NMN (an NAD^+ precursor). This indicates that 5-deazaflavin can enhance neuronal energy metabolism and potentially delay neurodegenerative processes. Furthermore, TND-1128 is being explored for anti-aging benefits; by boosting mitophagy (the cleanup of damaged mitochondria) and supporting cellular NADH/NAD^+ balance, it may protect tissues from age-related decline. While not a natural vitamin, 5-deazaflavin represents a novel class of redox-active molecules with therapeutic promise in conditions of metabolic and oxidative stress.
