Urolithin A

Description

Urolithin A is a microbiome-derived metabolite produced in the gut by the transformation of ellagitannins and ellagic acid (found in pomegranate, berries, etc.). Chemically, it is a dibenzo-α-pyrone structure with two hydroxyl groups. Urolithin A has garnered attention for its ability to induce mitophagy, the selective recycling of damaged mitochondria, which is a key mechanism in cellular aging. Preclinical studies demonstrated that Urolithin A supplementation improved mitochondrial function and muscle health: in aged rodents, it increased endurance and exercise capacity by enhancing mitochondrial respiration and upregulating genes involved in autophagy and muscle function. Urolithin A also shows anti-inflammatory effects and neuroprotective potential. For instance, it can cross the blood-brain barrier and has reduced neuroinflammation and improved memory in animal models of Alzheimer’s disease, partly by clearing dysfunctional mitochondria. In human trials, Urolithin A (commercially formulated as a supplement) was found to be safe and improved muscle strength and exercise performance in middle-aged adults over a 4-month period. By boosting NAD^+ levels and mitochondrial biogenesis, Urolithin A emerges as a promising anti-aging compound. Ongoing research is examining its benefits for muscle disorders, metabolic health, and longevity.
