Pregnenolone

Pregnenolone is a naturally occurring steroid hormone and is the precursor from which most other steroid hormones are synthesized (often called the “mother hormone”). It is biochemically derived from cholesterol and converted via enzymatic steps into progesterone, mineralocorticoids, glucocorticoids, androgens, and estrogens. Pregnenolone itself also functions as a neurosteroid in the central nervous system. It can modulate synaptic transmission, enhance NMDA receptor function, and influence memory and mood. Some studies in the 1990s suggested pregnenolone may improve cognitive performance and have antidepressant or anxiolytic effects, possibly by potentiating GABA_A receptor responses or through downstream neurosteroid metabolites. Clinically, pregnenolone levels decline with age similar to DHEA. It has been investigated in small trials for conditions like schizophrenia (where it showed some cognitive and negative symptom improvement) and for inflammatory diseases due to its immunomodulatory properties. Pregnenolone is available as an over-the-counter supplement in certain countries, touted for memory enhancement, stress reduction, and fatigue reduction. However, robust clinical evidence for these uses is limited. As a steroid precursor, if taken exogenously, it may theoretically shift the balance of downstream hormones, so caution and medical supervision are advisable for high doses.
