N-Acetylneuraminic Acid (Sialic Acid)

Description

N-Acetylneuraminic acid (Neu5Ac) is the predominant form of sialic acid in humans, a nine-carbon sugar acid. It is a crucial component of glycoproteins and gangliosides, especially abundant in the brain and nervous system. Neu5Ac residues cap the ends of cell-surface glycans and play key roles in cell–cell recognition, viral/bacterial entry, and immune modulation. In human nutrition, Neu5Ac is notably present in high amounts in breast milk oligosaccharides, and it has been implicated in infant brain development and cognitive function. Studies suggest that dietary sialic acid supports learning and memory by promoting synaptic membrane formation and neural plasticity. As a supplement, N-acetylneuraminic acid is being explored for neurodevelopmental benefits and for supporting cognitive health in aging (given that brain sialic acid levels can affect neuronal communication). Neu5Ac also has roles in immune regulation – for example, sialylation of cell surfaces can transmit “self” signals to prevent autoimmune reactions, and many pathogens bind to sialic acid as an initial attachment point. Supplementing sialic acid may, in theory, bolster glycoprotein synthesis in tissues with high turnover (like gut mucins or blood cells), but more research is needed.
