Lemairamin (WGX-50)

Description

Lemairamin (also known by the code WGX-50 or GX-50) is a plant-derived amide compound isolated from Zanthoxylum species. It has attracted interest for its analgesic and anti-inflammatory properties. Lemairamin acts as an agonist of the α7 nicotinic acetylcholine receptor (α7nAChR) in the spinal cord. Through this mechanism, it was shown to reduce neuroinflammation in models of neuropathic pain and Alzheimer’s disease. In animal studies, Lemairamin administration alleviated pain hypersensitivity: it dose-dependently inhibited chronic tonic pain in formalin tests and prevented mechanical allodynia (pain from normally non-painful stimuli) in neuropathic pain models. The analgesic effects were reversed by blocking α7nAChRs, confirming receptor-specific action. Additionally, Lemairamin increased the release of anti-inflammatory cytokine IL-10 and endogenous opioid β-endorphin in the spinal cord, suggesting it engages an immune modulatory and endorphin-mediated pathway to suppress pain. Early research also indicates Lemairamin has neuroprotective potential; by activating α7nAChRs, it may reduce microglial activation and thereby lower neuroinflammation associated with neurodegeneration. These findings position Lemairamin as a novel analgesic and neuroinflammatory modulator under investigation.
